Ion channels and G protein-coupled receptors as targets for invertebrate pest control: from past challenges to practical insecticides

چکیده

ABSTRACT In the late 1970s, we discovered that toxic bicyclic phosphates inhibit generation of miniature inhibitory junction potentials, implying their antagonism ?-aminobutyric acid (GABA) receptors (GABARs; GABA-gated chloride channels). This unique mode action provided a strong incentive for our research on GABARs in later years. Furthermore, minor structural changes conferred insect GABAR selectivity to this class compounds, convincing us possibility as targets insecticides. Forty years later, third-generation insecticides acting allosteric modulator antagonists at distinctive site were developed. G protein-coupled (GPCRs) are also promising pest control. We characterized phenolamine functionally and pharmacologically. Of tested receptors, ?-adrenergic-like octopamine revealed be most sensitive acaricide/insecticide amitraz. Given presence multiple sites action, ion channels GPCRs remain potential invertebrate

G-protein-coupled receptors as signaling targets for antiplatelet therapy.

Platelet G protein-coupled receptors (GPCRs) initiate and reinforce platelet activation and thrombus formation. The clinical utility of antagonists of the P2Y(12) receptor for ADP suggests that other GPCRs and their intracellular signaling pathways may represent viable targets for novel antiplatelet agents. For example, thrombin stimulation of platelets is mediated by 2 protease-activated recep...

Adhesion G Protein-Coupled Receptors as Drug Targets.

The adhesion G protein-coupled receptors (aGPCRs) are an evolutionarily ancient family of receptors that play key roles in many different physiological processes. These receptors are notable for their exceptionally long ectodomains, which span several hundred to several thousand amino acids and contain various adhesion-related domains, as well as a GPCR autoproteolysis-inducing (GAIN) domain. T...

G protein-coupled receptors as direct targets of inhaled anesthetics.

The molecular pharmacology of inhalational anesthetics remains poorly understood. Despite accumulating evidence suggesting that neuronal membrane proteins are potential targets of inhaled anesthetics, most currently favored membrane protein targets lack any direct evidence for anesthetic binding. We report herein the location of the binding site for the inhaled anesthetic halothane at the amino...

IUPHAR-DB: the IUPHAR database of G protein-coupled receptors and ion channels

The IUPHAR database (IUPHAR-DB) integrates peer-reviewed pharmacological, chemical, genetic, functional and anatomical information on the 354 nonsensory G protein-coupled receptors (GPCRs), 71 ligand-gated ion channel subunits and 141 voltage-gated-like ion channel subunits encoded by the human, rat and mouse genomes. These genes represent the targets of approximately one-third of currently app...
